Restart computer.
Does mouse/keyboard work plugged into a known working USB port on same computer.
Does mouse/keyboard work connected to a known working USB port on another computer.
Does a replacement mouse/keyboard work connected to a known working USB port on same computer
Does a replacement mouse/keyboard work connected to a known working USB port on another computer.
Has software been installed for a 3rd party mouse or keyboard?
Does that software need update or rollback?
Check compatibility with OS X version.
Remove vendor software, and test, often the native OS X can accommodate for a 3rd party mouse/keyboard without vendor software.
